Hornchurch new boy Stefan Payne struck twice to help seal the points in south London.
Payne, who joined the club from Ebbsfleet late last week, netted two goals in 11 minutes, after replacing Carl Rook on the hour, to put Urchins 3-1 up.
And an injury-time penalty for the hosts could not deny Jimmy McFarlane’s men a memorable win.
Urchins took time to settle and Dulwich saw a Kevin James free-kick punched clear by a diving Inigo Echepare.
George Purcell’s first-time shot was tipped wide by Phil Wilson at the other end, before Phil Starkey fired just wide from the corner.
Mathieu Boyer was off-target for Dulwich before Urchins took the lead on 21 minutes as Purcell hammered home from range.
Purcell and Ricky Hayles went close to a second before Nyren Clunis levelled in the 33rd minute and defences remained on top before the break.
Alex Bentley forced Wilson into action after the restart and Elliot Styles saw a header tipped over from a Derek Duncan free-kick.
McFarlane made two changes around the midway mark of the second half and it paid off as Tambeson Eyong won possession and began a move which ended with Payne dispossessing Peter Adeniyi and stroking the ball home.
Styles denied Dulwich a reply, with the home fans appealing for a penalty, and Urchins added their third when Purcell sent Payne through to score again.
Ahmed Deen and Ryan James wasted good chances for the home side, before Echepare denied James with a stunning save.
Dulwich were awarded a late penalty, which Erhun Oztumer converted, but it was too late to preserve their unbeaten record as Urchins returned home with the spoils.
